Course objectives:
|
The subject deals with basic immune mechanisms under physiological conditions and in immunopathological conditions and the principles of immunological examination methods with the aim of equipping students with the ability to examine the humoral and cellular components of the immune system. Teaching takes place in the laboratories and seminar room of the Institute of Immunology and Allergology of the Pilsen University Hospital. The final examination includes topics discussed in the subjects Immunology 1 and Immunology 2.

1. Th1/Th2 and Tc reactivity - repetition.
2. Defense against extra and intracellular pathogens.
3. Defense against viruses and defense against multicellular parasites.
4. Antitumor immunity.
5. Transplant immunity.
6. Immunological relationship of mother and fetus, Rh incompatibility.
7. Immunopathological reactions of type I. - IV.
8. Immunodeficiency. Autoimmunity.
9. Allergies
10. Therapeutic interventions in the immune system.
11. Immunotherapy. New trends in immunological and allergological diagnostics.
A Lab exercise
1. Introduction to the evaluation of autoantibodies using NIF.
2. Determination of ANA antibodies using the indirect fluorescence method.
3. Examination before bone marrow transplantation.
4. Special methods - density gradient separation, MTT - test.
5. Special methods - cross match test.
6. Special methods - NK cell tests.
7. Special methods - determination of intracellular production of cytokines.
8. Determination of specific IgE - immuno cap.
9. Determination of specific IgE - component analysis, ISAC.
10. Repetition of summer semester topics.

to define basic immunological terms and characterize individual components of non-specific and specific immunity |
to explain the principles of laboratory methods for monitoring humoral and cellular immunity |
Skills - students are expected to possess the following skills before the course commences to finish it successfully: |
to observe the principles of work safety and fire protection in the immunological laboratory |
to perform protein detection under supervision, including Ig nephelometry and turbidimetry and detection of circulating immunocomplexes |
using the flow cytometry method to determine under supervision T and B lymphocyte phenotyping, basophil activation test, lymphocyte blast transformation and oxidative burst |
to master the principle of ELISA immunochemical methodology using an enzyme to mark an antigen or an antibody |

characterizes individual types of defense mechanisms against extra- and intracellular pathogens, viruses and multicellular parasites |
explains the role of individual components of immunity in the defense against tumors and the specifics of transplant immunity, |
describes the mechanism and interprets the possible consequences of Rh incompatibility between mother and fetus |
characterizes individual types of immunopathological reactions I. - IV. |

prepares the specimen for reading under a fluorescence microscope |
performs a cross match test to select the most suitable donor-recipient pair before transplantation |
handles the basics of special methodologies - MTT-test, NK cell tests, determination of intracellular cytokine production |
according to the protocol, he performs the determination of specific IgE |
